SCREENING SYSTEMS
Physica
has set up a unique screening system which accelerates the process of
formulation selection. This selection method is based on the identification
of several physical and biological parameters.
This approach can be applied on compounds resulting from drug discovery
and which can benefit from a "physical optimization". It can
also be used for molecules in later stages of development or already marketed.
Physical
screening system
Physica has developed an efficient "physical" screening tool
based on measurements of parameters such as log P, log D, and artificial
membrane partition model. This pre-formulation system can be automated
and uses only few milligrams of a drug. During this phase, Physica generates
data on key compound properties and selects drug delivery agents with
the aim to optimise drug's membrane diffusion properties.
Biological screening
system
Physica has developed in vitro cell systems which are used to simulate
drug transport across biological barriers. Here, the formulation prototypes
are screened using biological membranes, thus permitting the detection
of the most promising candidates for pre-clinical trials .
Gastrointestinal
Absorption
Physica
uses a model of the intestinal mucosa based on the human colon carcinoma
cell line Caco-2.
These cell culture models allow the permeability and metabolic processes
to be examined rapidly under controlled conditions.
Pulmonary
absorption
In
the same manner, PHYSICA has developed pulmonary cell models based on
human pulmonary cell line Calu 3.
